ABSTRACT
OBJECTIVE@#To construct expression vector for the SEA-EGF fusion gene.@*METHOD@#Clone the SEA gene and the EGF gene segment with PCR and RT-PCR independently, and connect this two genes by the bridge PCR. Insert the fusion gene EGF-SEA into the expression vector PET-44. Induced the secretion of the fusion protein SEA-EGF by the antileptic.@*RESULT@#The gene fragment encoding EGF and SEA mature peptide was successfully cloned. The fusion gene EGF-SEA was successfully constructed and was inserted into expression vector.@*CONCLUSION@#The new recombinant expression vector for fusion gene EGF-SEA is specific for head and neck cancer, laid the foundation for the further study of fusion protein SEA-EGF targeting immune therapy in head and neck tumors.

Objective To obtain the frequency weighting contour for seated Chinese young males in z axis.Method 10 male volunteers served as the subjects.They were exposed to standard vibration stimulus (a sinusoidal vibration of 1.0 m/s2 at 8 Hz) and 14 kinds of sinusoidal experimental vibration stimuli at frequencies from 4 Hz to 80 Hz.The magnitude of the experimental vibration stimulus was changed by the subject through a signal condition and switching device made by ourselves to obtain an equal vibration sensation to the standard vibration.The data were fitted to the same form of the frequency weighting Wk of ISO 2631-1 with the least square method and optimization of parameters.Result Compared with ISO 2631-1,the frequencies weightings of Chinese young males at 16～80 Hz are significantly higher than those of ISO 2631-1(P<0.005),but there are no significant difference between two frequency weighting contours at other frequencies. Conclusion Chinese young males are more sensitive to vibration at 16～80 Hz in Z axis.
ABSTRACT
Aim To investigate the effects of lipophilic fraction of Ganoderma lucidum on PC12 differentiation. Methods Rat PC12 cells were cultured in DMEM medium. Differentiation of PC12 cells was evaluated by monitoring neurite outgrouth of PC12 cells with phase-contrast microscopy after treatment with lipophilic extracts of Ganoderma lucidum or it's effective component B_2. Expression of neuronal marker,microtubule-associated protein-2,was determined with immonofluorescence and western blot analysis. Results Lipophilic extracts of Ganoderma at the concentrations between 125 and 500 mg?L -1 induced neurite outgrowth of PC12 cells in a dose-dependent manner. Nearly all cells showed morphological differentiation in response to lipophilic extracts of Ganoderma. Lipophilic extracts of Ganoderma also potentiated NGF-induced differenti-ation of PC12 cells. Lipophilic extracts of Ganoderma increased the expression of MAP-2. Conclusion Lipophilic extracts of Ganoderma lucidum can induce differentiation of PC12 cells and potentiate NGFs actions.
